Introduction: One of the novel approaches in stress management and health improvement in aquatic organisms is the use of dietary supplements with immune-boosting properties. Purple coneflower, Echinacea purpurea, as one of the medicinal plants, due to its phenolic compounds, glycosides, alkaloids, and antioxidant, anti-inflammatory, and immune-boosting properties, can be used as a natural supplement to improve immune system performance and resistance against environmental stresses in aquaculture. Research in aquatic species has shown that the use of medicinal plant including E. purpurea can have positive effects on the physiological status, immunity, and health of aquatic organisms when faced with environmental stresses and diseases. In this study, the effects of a diet containing purple coneflower extract and powder on hematological and immune-antioxidant factors of the hemolymph of Pacific white shrimp (Litopenaeus vannamei) under pH stress were investigated.
Materials and Methods: Shrimp were fed with diets containing different concentrations of purple coneflower extract (0.25%, 0.5%, and 1%) and powder (0.25%, 5% and 10%) for 68 days. Subsequently, the performance of blood and immune-antioxidant factors in the hemolymph, including total and differential hemocyte counts and the activity of enzymes such as lysozyme, superoxide dismutase, glutathione peroxidase, and catalase were evaluated after exposure to acidic (pH 4.5) and alkaline (pH 9.5) stress conditions for 24 hours.
Results and Discussion: Under acidic and alkaline pH conditions, the total hemocyte count was significantly lower than in the control group in different treatments except for treatment containing 0.25% extract of purple coneflower (P<0.05). The large granular cells did not show significant difference among treatments in response to acidic pH stress (P>0.05), however, they were significantly lower than the control group in different treatments except for treatment containing 0.25% extract of purple coneflower in response to alkaline pH stress (P<0.05). Additionally, the results showed that under acidic and alkaline pH stress conditions, the semigranular and hyaline cells and the activity of lysozyme, superoxide dismutase, glutathione peroxidase, and catalase, did not show significant differences compared to the control group at lower doses of extract and higher doses of purple coneflower (P>0.05). However, with an increase in extract dosage and a decrease in powder dosage, the immune-antioxidant factors in the shrimp's hemolymph were reduced compared to the control group.
The results of this study indicate that purple coneflower, at certain concentrations, led to changes in the hemolymph immune-antioxidant response under acidic and alkaline pH conditions, resulting in a reduction in hemocyte counts and then activity of immune-antioxidant enzymes. The results showed that the plant, at some doses, was not effective in regulating the immune-antioxidant response of shrimp under acidic and alkaline pH conditions.
Conclusion: The results of this study indicated that different concentrations of purple coneflower extract and powder can affect the immune-antioxidant system of shrimp in response to pH stress. The treatment with 0.25% the purple coneflower extract exhibited the most positive effect on shrimp immunity in response to pH stress, where hemocyte counts and immune-antioxidant enzyme activity in this treatment did not differ significantly from the control group under both acidic and alkaline pH stress. This positive effect is likely due to the presence of active metabolites, especially active polyphenolic compounds and polysaccharides, in the purple coneflower extract. When using the purple coneflower as a natural supplement in shrimp feeding, their concentrations should be carefully managed to prevent negative impacts on the immune system.

Introduction: Sturgeon aquaculture is of great economic importance due to the high value of caviar and meat. However, challenges such as reproductive and growth limitations in female broodstock hinder productivity in controlled environments. Steroid hormones are among the key factors regulating growth and reproduction in fish. Among them, 17β-estradiol (E2) plays a crucial role in sexual differentiation, gonadal development, and metabolic regulation. Slow-release hormone delivery methods have been proposed as a potential approach to enhance growth and maturation in fish species. The aim of the present study was to investigate the effect of slow-release E2 injection on the growth performance of female Siberian sturgeon, Acipenser baerii at different sexual maturity stages.
Materials and Methods: A total of 30 female Siberian sturgeon were selected and categorized into three groups based on their sexual maturity stage (II, III, and IV). Each stage was divided into two subgroups: 1) E2-treated group (received 2 mg E2/kg body weight); 2) Control group (received 0.2 mL coconut oil/kg body weight). The hormone was administered intraperitoneally every two months, and the experiment lasted four months. Biometric measurements and growth performance, including body weight, total length, weight gain (WG), specific growth rate (SGR), body weight increase (BWI), condition factor (CF) and average daily growth (ADG) were conducted at 30-day intervals. Statistical analysis was performed using repeated measures analysis of variance to assess the effects of treatment, maturity stage, and time on growth indices.
Results and Discussion: The findings revealed a significant interaction (P<0.05) between treatment, sexual maturity stage, and time in all growth parameters except CF. The highest final body weight (7950 ± 320.93 g) and total length (120.60 ± 11.20 cm) were recorded in the E2-treated fish at stage IV after 120 days, while the lowest body weight (4170 ± 137.47 g) and total length (94.50 ± 0.93 cm) were observed in control fish at stage II after 30 days (P<0.05). The highest WG (610 ± 18.70 g), SGR (0.42 ± 0.00 %/day), BWI (13.62 ± 2.33 %), and ADG (20.33 ± 3.30 g/day) were recorded in the E2-treated fish at stage II after 90 days (P<0.05). The CF remained unchanged across all groups, and no mortality was observed throughout the experiment. These findings indicate that E2 administration promotes somatic growth at early maturity stages (II), whereas at advanced stages (IV), it facilitates gonadal development. The hormonal influence on growth appears to be stage-dependent, with higher energy allocation towards reproductive tissues at later stages. Similar studies on other sturgeon species (e.g., Huso huso, and Acipenser stellatus) suggest that E2 stimulates insulin-like growth factor-I production and enhances protein synthesis, ultimately improving growth. However, excessive doses may negatively impact somatic growth by favoring gonadal tissue development.
Conclusion: Slow-release E2 injection significantly affects the growth performance of female Siberian sturgeon, with stage-dependent effects. At early maturity stages (II), it stimulates somatic growth, while at advanced maturity stages (IV), it promotes reproductive development. These findings suggest that timing and dosage optimization are crucial for maximizing the benefits of hormonal treatments in sturgeon aquaculture. Further research is needed to explore the molecular mechanisms underlying E2's role in growth regulation and to determine the long-term reproductive consequences of hormone administration.

Introduction: Synbiotics are combination of prebiotics and probiotics that are used in the aquaculture industry, but the mechanism of action of synbiotics can be different as growth and immunity stimulants in aquatic animals. In general, synbiotics optimize nutrient metabolism and create favorable biological conditions by maintaining the dominance of probiotic bacteria in the intestinal microbiota through the consumption of non-digestible prebiotics, thereby improving growth and immune performance. This study aimed to determine the effects of synbiotic supplementation (mixture of probiotic Bacibus subtilis and prebiotic inulin) on growth performance, digestive enzymes, biochemical factors, and resistance to Aeromonas hydrophila in Nile tilapia (Oreochromis niloticus).
Materials and methods: Two hundred and forty fish with initial body weight of 5.18 ± 0.83 g were randomly distributed in 12 tanks with 60 L of water. Fish were randomly fed with 4 experimental treatments (each with 3 replicates) with different levels of synbiotic supplement diets for 56 days, as follows: BI0: control without supplement addition, BI1: (2.5 × 1011 CFU/g probiotic B. subtilis + 2.5 g inulin), BI2: (2.5 × 1011 CFU/g probiotic B. subtilis + 5 g inulin), BI3: (2.5 × 1011 CFU/g probiotic B. subtilis + 10 g inulin).
Results: The results of this experiment showed that the highest weight and lowest feed conversion ratio were obtained in BI2 and BI3 compared to BI1 and the control. Digestive enzyme assays showed that there was no significant difference in the lipase and amylase activity between the synbiotic and control treatments, while protease enzyme increased significantly in the synbiotic feeding treatments. There was no significant difference in the concentration of protein, alanine aminotransferase and aspartate aminotransferase between the synbiotic feeding treatments and the control. The antibacterial activity of blood plasma of tilapia fed BI3 was significant compared to the BI0 (control).
Discussion and Conclusion: Synbiotics can improve the general health of aquatic animals, but it is very important to determine the effective compounds and doses on growth performance, digestive enzyme secretions, and blood biochemical components. Therefore, in the present study, a synbiotic combination (probiotic subtilis with different amounts of prebiotic inulin) in nutritional treatments with a dose of 2.5 × 1011 CFU/g probiotic B. subtilis + 5 and 10 g inulinof diet is recommended for Nile tilapia. Overall, growth performance, digestive secretions, immunity, and resistance to Aeromonas hydrophila in tilapia fed with different levels of synbiotic (probiotic B. subtilis + prebiotic inulin) improved in the experimental treatments, especially BI2 and BI3.

Introduction: The primary operational cost in aquaculture is attributed to feed expenses, which plays a critical role in growth enhancement, immune system performance, and disease resistance. To address this, various strategies have been proposed for feed management, including short-term or long-term fasting, as well as cyclic periods of fasting and refeeding to achieve compensatory growth. Another strategy involves restricting the protein content in the diet within a specific timeframe. This method focuses solely on limiting the protein content in the fish diet for a specific duration, after which the protein level is restored to an optimal and balanced level without interrupting the feeding process. Therefore, formulating diets that maximize growth while conserving protein resources, considering economic viability, and minimizing adverse environmental effects, is essential.
Material Methods: A total of 270 red tilapia, with an average initial weight of 5 ± 0.13 g were randomly distributed among 18 aquaria, each with a capacity of 100 liters (15 fish per tank). The fish were subjected to various protein-level dietary treatments over 8 weeks and were fed three times daily at 9:00, 13:00, and 16:00 until satiation. The experimental treatments were as follows; control group: feeding with a diet containing 38% optimal protein, treatment 1: feeding with a diet containing 32% protein restriction, treatment 2: feeding alternately every other day with a diet containing 32% protein restriction and a diet containing 38% protein, treatment 3: feeding alternately every week with a diet containing 32% protein restriction and a diet containing 38% protein, treatment 4: feeding a 32% protein diet for the first 3 weeks, followed by a 38% protein diet for the remaining 5 weeks, and treatment 5: feeding a 32% protein diet for the first 4 weeks, followed by a 38% protein diet for the following 4 weeks. At the end of the experiment, growth, nutritional indices, and body composition were assayed. Enzyme activity was assessed using specific substrates and diagnostic kits. Subsequently, a One-way analysis of variance (ANOVA) was employed to compare the differences among the treatments, and the significance of the results was determined using Duncan's multiple-range test.
Results and discussion: The findings of this study revealed that the control group, which was fed a diet containing 38% optimal protein throughout the experimental period, achieved the highest growth performance and the best feed conversion ratio (FCR). Interestingly, treatment 4, which involved feeding the fish with a 32% protein-restricted diet for the first 3 weeks and a 38% optimal protein diet for the remaining 5 weeks, showed no significant difference in growth performance and FCR compared to the control group (P>0.05). This suggests that when refeeding with an optimal protein diet, moderate protein restriction can induce compensatory growth without negatively impacting growth performance or feed efficiency. Regarding body composition, the control group exhibited the highest protein and lowest lipid content in the carcass. However, treatment 4 showed no significant difference in these indices compared to the control group (P>0.05), indicating that the fish could maintain their body composition despite the initial protein restriction. Similarly, the lack of significant differences in moisture and ash content among the experimental groups further supports the idea that moderate protein restriction does not adversely affect the overall nutritional quality of the fish. The activity of digestive enzymes, including total protease, trypsin, chymotrypsin, and lipase, was the highest in the control group. However, treatment 4 showed no significant difference in digestive enzyme activity compared to the control group (P>0.05), suggesting that the fish were able to maintain their digestive efficiency during the refeeding period. This is a critical finding, as digestive enzyme activity is a key indicator of nutrient absorption and utilization in fish.
Conclusion: In conclusion, the results of this study demonstrate that moderate protein (3 weeks) restriction, when followed by adequate refeeding (5 weeks), can induce compensatory growth in red tilapia without negatively impacting growth performance, body composition, or digestive enzyme activity.

Introduction: Aquaculture is the fastest-growing source of animal protein globally and currently provides nearly half of the world's seafood consumption. Due to the limited fishery resources from ocean catches, this shortfall must be addressed through the aquaculture industry. To tackle the issues of fishmeal shortages in recent decades, special attention has been given to the use of alternatives such as insects in aquaculture feed. Insects are one of the most diverse animal groups in the world, with some species possessing suitable nutritional value. Among insects, the yellow mealworm Tenebrio molitor stands out due to its high protein and fat contents. It also contains considerable amounts of bioactive compounds and chitin with antioxidant potential, antibacterial peptides, and other immune stimulants that enhance immunity and improves hematological parameters in various aquatic species. In this study, the effects of replacing fishmeal with the yellow mealworm larvae meal were investigated on blood parameters, serum biochemical composition, and liver enzymes in Beluga, Huso huso.
Materials and methods: A total of 360 fish with an average weight of approximate 112.9 ± 8.0 g were randomly distributed to polyethylene circular tanks with a capacity of 1500 liters. The experimental treatments (each with three replicates) included the replacement of 0% (control), 20%, 40%, 60%, 80%, and 100% of yellow larval mealworm (MW) meal with fish meal. At the end of the trial, blood sampling was performed using a syringe from the fish's caudal fin. The counting of red blood cells and white blood cells was done using a hemocytometer (Neubauer), and the hematocrit was measured through the microhematocrit method, while the hemoglobin concentration was measured by the cyanmethemoglobin method using a spectrophotometer at a wavelength of 540 nm in grams per deciliter. Liver enzyme levels including alkaline phosphatase (ALP), aspartate aminotransferase (AST), and alanine aminotransferase (ALT) were measured using the α-ketoglutarate method. Lactate dehydrogenase (LDH) was tested using the lactate-pyruvate method to determine the possibility of tissue damage. Data analysis was performed using One-Way ANOVA and Duncan's multiple range comparison tests were performed at a significance level of P<0.05.
Results: According to the results of growth indices, the highest body weight gain and specific growth rate were achieved in treatments substituted with 40% and 60% MW (P<0.05). The survival rate of the fish did not show a significant difference between the treatments (P>0.05). The number of white blood cells, red blood cells, hemoglobin, and hematocrit decreased as mealworm powder increased in diet, with the lowest values observed in 100% MW (P>0.05). The values of MCH and MCHC significantly decreased with 100% MW replacement (P>0.05). Lymphocyte numbers were significantly higher in 40% and 60% MW groups compared to the 20% (P>0.05). The values of albumin and total protein improved with dietary replacement of MW powder, except for the 100% replacement (P<0.05). The concentration of triglycerides in group with 20% MW was similar to that of the control (P<0.05); however, it decreased as the MW replacement increased (P<0.05). Hepatic enzymes such as AST, ALT, and ALP decreased with the inclusion of MW in the diet (P<0.05).
Discussion: Total protein and albumin play an important role in immune responses, and the improvement of these parameters in the current study may indicate enhanced immunity in beluga after replacing fishmeal with MW. Additionally, diets containing insect meal improve the microbial composition of the digestive system, ultimately affecting fish immunity and health. Chitin also regulates fat metabolism by preventing the hepatic circulation of bile acids and interfering with the natural digestion and absorption of fats in the intestine and the biosynthesis of fatty acids in the liver.
Conclusion: Based on the results, it is not feasible to completely replace MW meal with fishmeal, and a 20% replacement level is suggested as the optimal for beluga. However, substitution levels up to 40-60% may also be considered with caution.

Introduction: The Kutum (Rutilus frisii), a member of the Cyprinidae family, is a unique species of bony fish native to the Iranian shores of the Caspian Sea. It has significant potential as a suitable candidate for introduction to aquaculture industry in Iran. Stocking density is a critical factor in fish pond management, as it often negatively impacts fish growth and welfare in aquatic environments. The negative impact of stocking density on growth performance is primarily due to the induction of stress in fish, leading to reduced feeding and growth rates. Among the most significant reasons for the occurrence of stress in this context are the interactions between fish, competition for food resources, and the habitat required. Therefore, determining the optimal stocking density for Kutum during the grow-out phase in earthen ponds is essential for enhancing productivity and maximizing economic profitability.
Materials and Methods: In this research, the growth performance and survival rate of Kutum were evaluated in the grow-out stage with an average weight of 60.89 ± 7.21 g in two densities of 5600 and 8000 fish per hectare in 700 m2 earthen ponds during the 3-month period. The fish were fed to satiation with extruded feed twice daily at 9:00 AM and 1:00 PM. The physical and chemical parameters of the water, including temperature, dissolved oxygen, nitrite, nitrate, ammonia, phosphate, and bicarbonate were measured using the AOAC (2023) method. Plankton and benthos sampling and identifying were conducted seasonally during summer and autumn from earthen ponds based on standard methods. To evaluate growth trends based on experimental treatments, Kutum were biometrically assessed. The length and weight of 50 fish were measured each time using a biometric board and a scale with an accuracy of 0.01 g. The growth indices and survival rate were calculated by reference formulas. For data analysis, One-way analysis of variance (ANOVA) was used. Additionally, Duncan's multiple range test was employed to examine the significance of mean differences among experimental treatments. Data evaluation was performed using SPSS software.
Results and Discussion: The results showed that the growth performance of Kutum decreased significantly by the elevation of density (P≤0.01). The average percentages of weight gain in the first and second treatments were 248.62 ± 102.04 and 115.58 ± 49.51 g and the average food conversion ratio were 2.15 ± 0.92 and 4.23 ± 1.64, respectively, and the difference between the treatments was significant (P≤0.01). Also, the difference between the 1st and 2nd treatments in the retention rate of the reared fish at the end of the rearing period was not significant and it was 80.58 ± 2.43 and 76.07 ± 3.51, respectively (P>0.01). The amount of final fish crop in treatment 1 was higher than in treatment 2; 1646.51 ± 109.55 and 1589.91 ± 88.94 kg/ha, respectively, but the difference between the treatments was not significant. The results of our study showed that the average data for the earthen ponds during the farming period fell within the standard range for warm-water fish, and this indicates the suitable condition of the measured parameters during various stages of farming throughout the production period. Higher stocking density led to a reduction in the diversity and abundance of live food (benthos and zooplankton) in the pond due to faster consumption by the fish within a shorter period during the farming cycle. Our study has shown that fish at higher densities are exposed to increased competition for food and space, leading to physiological stress. This can resulted in reduced growth indices, feeding efficiency, feed conversion efficiency, product yield, and survival rates. The results of our study align with findings from other researchers regarding the effects of stocking density on the performance of other aquatic species.
Conclusion: In general, this study concluded that high stocking density has significant effects on the growth, survival, and production yield of Kutum during the grow-out stage. Therefore, it is recommend to stock 5,600 Kutum per hectare in 700-m2 earthen ponds to achieve higher production with optimal performance.
