ارزیابی خواص ضدباکتریایی عصاره الکلی و اتری جلبک Scenedesmus dimorphus بر گونه-های باکتری‌های Micrococcus luteus و Aeromonas hydrophila

چکیده

 هدف از این مطالعه تعیین خواص ضد­باکتریایی موجود در عصاره­های الکلی و اتری (دی­اتیل­اتر و متانولی) جلبکScenedesmus dimorphusعلیه باکتری گرم مثبت  Micrococcus luteusو باکتری گرم منفیAeromonas hydrophila بود. این جلبک به صورت خالص کشت داده شده و پس از تأمین حجم مناسب جلبک، عصاره­های دی­اتیل­اتر و متانولی آن با استفاده از روش انتشار در آگار استخراج گردید. ارزیابی خاصیت ضدباکتریایی با استفاده از روش چاهک انجام شد. در این مطالعه از دو تیمار دی­اتیل اتر و متانول با سه تکرار استفاده شد. نتایج به دست آمده نشان داد که عصاره متانولی جلبک S. dimorphus (50، 100 و 200 میلی­گرم در لیتر) دارای خاصیت مهار­کنندگی علیه باکتری­های M. luteus وA. hydrophila  با هاله عدم رشد به ترتیب در دامنه 11-13 و 13-21 میلیمتر بود. در مورد عصاره دی­اتیل ­اتر (50، 100 و 200 میلی­گرم در لیتر) نیز هاله عدم رشد در دامنه 7/23-9 و 3/6-6/21 میلیمتر مشاهده شد. برای شناسایی ترکیبات موجود در عصاره­ها گاز- کروماتوگرافی عصاره­های متانولی و دی­اتیل­اتری انجام شد و نتایج نشان داد که بیشترین ترکیبات در عصاره­های متانولی و دی­اتیل­اتری، هیدروکربن­­ها و استرها با غلظت مؤثر 200 بودند. عصاره اتری بر باکتری گرم مثبت و عصاره متانولی بر باکتری گرم منفی بیشترین تأثیر را داشت. این نتایج حضور ترکیبات ضد باکتریایی را علیه این دو گونه باکتری در جلبک S. dimorphus نشان می­دهد.

Antimicrobial properties of diethyl ether and methanol extracts of algae Scenedesmus dimorphus against Gram positive bacterium Micrococcus lutesus and Gram negative becterium, Aeromonas hydrophila were evaluated. The algae was pure cultured and once the required volume and number of cells achieved coli using agar well diffusion method diethylether and hexane extracts of the algae cells were extracted. Antibacterial properties of the extracts were evaluated using well diffusion method. The results showed inhibitory activities of methanol and ether extracts of Scenedesmus dimorphus against M. luteus and A. hydrophila with 11-13 mm and 13-21 mm diameters of inhibition zone, respectively. In case of diethyl ether extracts also the inhibitory activity observed with diamaters of inhibition zone in the range of 9-23.7 mm and 21.6-6.3 mm respectively against M. luteus and A. hydrophila. Gas chromatography mass spectrophotometry was used to identify different substances within diethyl ether and methanol extracts with hydrocarbon and ester with highest perentage among other substances. The results confirmed antimicrobial activities of extracts from S. dimorphus against bacteria used in this study.
